Auto Save
This is a MUST HAVE feature for those of us who paint all day without saving sometimes. Hey it happens. I can be away from my computer and something happens while I am gone

Additional texture mapping options
Currently the only texture mapping option is UV, and it would be very helpful if we had access to some other types. Cube projection mapping would be particularly useful when applying textures to models with visible seams.

invert mask color by holding ctrl or alt.
I am often have to invert my color from white to black - it would be cool to have some option for quick invert during editing

when I zoom in using the middle scroll it should use the mouse location as the center point.

The other way (the way it does it now) seems to make no sense.
Zoom, move around a bit, Zoom more, bit more fiddly moving.
Finally get the thing you want.
Easy to improve.
I can teach a kid. "HOLD ALT.", "SCROLL TO THING YOU WANT BY POINTING AT IT" I can not teach them the other way, it's too infuriatingly counter-intuitive, and harms productivity, thus is non-rewarding. Tough to teach.

Vray and Arnold Shader
Vray and Arnold Shader: This would facilitate the workflow a lot.
Because the export of the textures and replace in Arnold or VRay needs a lot of adjustments in the shader itself.

Ability to choose a layer as an image input for a Substance.
That would allow for instance to create advanced substance materials, with effects driven in real time by painting on a layer (that is set as an image input in the substance).

Additional shaders
Need more shader options in Substance Painter including:
Phong, Cook Torrance, Beckman, Blinn, Flat, BRDF, Layered

Blur brush
For example simple holding shift to blur texture

Draw straight lines by holding a key
Similar to photoshop where you can paint straight lines by holding Shift

Allow for changing the default location of the materials
The location: Users***\Documents\Substance Painter\shelf\presets\materials
is no good for me.
I do everything from the D: drive other than run the OS.
Please allow us to change the Users***\Documents\Substance Painter\shelf\presets\materials location to one of our choosing?
There is no room on that drive for that.

Gradient tool
It`ll be nice to have a tool to add gradient (ex: black to white from bottom to top of the model), ideally linear and radial ones.

Ability to quickly copy and paste entire layer stacks from one textureset to another
I find it terribly frustrating not to be able to copy and paste entire layer stacks. and from one textureset to another you can't copy and paste anything atm. It would also be cool to be able to save a layerstack to an external preset file that you could load onto another scene entirely. That way if you have a lot of assets to texture that all share the same materials, you`d have to do one and get the rest pretty far really quickly. Simply open new object, load layer stack, base texturing done. That way you can focus on…

Ability to see transparency in the viewport
When painting opacity mask, it would be useful to see the transparency in the viewport.

Directional light and shadow support
Image based lighting is great, but it lacks a main directional light casting high quality shadows. It would help a lot in model showcasing.

Swap foreground and background color with shortcut
Simialr to photoshop's X shortcut. This would provide a fast way to swap black and white while painting masks.

selection copy/paste/move/rotate/scale
basically the selection and transform capabilities of photoshop - select an area, cut or copy, paste into same or different layer, use transform gizmo to adjust in either 2d or 3d, release selection.
working with many layers to achieve this is unwieldy.

Lazymouse and advanced Brush settings
A Feautre like Lazymouse would be helpful, with it it would be nice to see more brush options like straight lines, dotted lines etc.

set library paths in preferences like in Designer
I like the way, Substance Designer lets you setup your library by defining paths in the preferences and filtering them.
In Painter, I always have to add the alphas manually per project...
